Recent breakthroughs in immunotherapy have significantly reduced cancer mortality worldwide. Maintaining this impactful progress has sparked significant interest and investment for researchers to further explore novel immunotherapeutic strategies.

A key hurdle hindering desired experimental outcomes revolves around effectively evaluating cell populations with high autofluorescence or low biomarker expression.

Cytek’s innovative Full Spectrum Profiling™ (FSP™) technology allows for simultaneous multiplex measurements of 40 colors or more from a single tube. Offering unprecedented flexibility and time savings, FSP technology transforms flow cytometry, allowing scientists to use a wide range of fluorochrome combinations.

This high-parameter approach significantly improves traditional technologies, minimizing the time and resources spent on multiple tubes, assays, and reagents while reducing variability in small sample measurements.

FSP technology resolves and differentiates rare, dim, and challenging populations. With the autofluorescence extraction feature, users can define autofluorescence as separate or multiple parameters for resolving complex and highly autofluorescent populations.

FSP technology measures and identifies close or overlapping fluorescent emissions spectra in multicolor panels which allow for predictable fluorochrome interactions and effective panel design. This leads to increased data resolution for rare and dim populations with minimal spillover correction. Cytek also simplifies experiment setup and prevents incompatible filter errors with one configuration for all applications. Together, these innovations can help to accelerate the design of personalized immunotherapies that modulate the immune response against cancer cells, all important for cancer immunotherapy and translational research, where a breakthrough can transform lives.

The Cytek® Aurora™ flow cytometer, which accommodates up to five lasers, three scatter channels, and 64 fluorescence channels, is powered by FSP technology and captures more detailed information from each fluorochrome regardless of the application complexity. Redefining sensitivity with state-of-the-art optics and low-noise electronics, the Cytek Aurora flow cytometer can detect low-abundance biomarkers and evaluate microparticles and extracellular vesicles (EVs) with high resolution and accuracy.

Powerful use cases of the Cytek Aurora flow cytometer include the Amsterdam University Medical Center and the Agency for Science, Technology, and Research in Singapore. Amsterdam University Medical Center has used the Cytek Aurora flow cytometer to quantify antigen-presenting cells and develop effective cancer vaccine delivery protocols. The Agency for Science, Technology, and Research has leveraged the Cytek Aurora flow cytometer to develop patient-specific, modified dendritic cells and monitor the effects of personalized cell therapy.

A testimonial by Dr. Ueno Hideki from Kyoto University emphasizes the impact of the Cytek Aurora analyzer. Specimen processing, staining, and data capture are similar to those of conventional flow cytometers, making it easy to transfer between instruments. Of note, the Cytek Aurora CS system was recently awarded the ‘Cell Sorting Product of the Year’ by BioTech Breakthrough. From over 1,500 nominations across 12 countries, it stood out as the only cell sorter on the market that combines FSP technology with high-performing cell sorting to help scientists better understand complex biological systems.

Another notable offering is the Cytek® Amnis® ImageStream®X Mk II imaging flow cytometer, which performs fluorescence analysis combined with single-cell imaging. Powerful artificial intelligence and machine learning-driven software seamlessly link images with quantitative data to define populations and cellular statuses. Users only need to click on a dot in any histogram to view corresponding images, click on a histogram bin to examine cells in the bin, and draw gates on dot plots and view the resulting populations to validate outcomes. The Cytek Amnis ImageStreamX Mk II system facilitates analysis of quantitative internalization, localization, cellular interactions, cellular exhaustion mechanisms, and cytotoxicity studies, opening new avenues for precise and insightful applications. In addition, for researchers focused on EVs, this instrument is highly sensitive and able to identify and resolve submicron particles.

Cytek also offers optimized reagents and kits for immunophenotyping and immune monitoring. Cytek’s cFluor® reagents and panels are useful for distinguishing and quantifying T, B, and NK cell subsets in whole blood and peripheral mononuclear cells. For translational research, optimized panels and kits have been developed to evaluate acute myeloid leukemia (AML), measurable residual disease (MRD), myeloid-derived suppressor cells (MDSCs), and natural killer (NK) cells to boost therapeutic discoveries. These panels can monitor anti-tumor activity during targeted antibody, chimeric antigen receptor (CAR), CAR-T, and CAR-NK therapy developments, paving the way for more cost-effective high-dimension experiments.

In addition to its more than 2,000 Cytek instruments deployed globally, Cytek has over 1,450 peer-reviewed publications featuring its FSP technology with applications across oncology, immunotherapy, drug and vaccine development, infectious diseases, and microparticle research.
